Our laboratory, utilizing next-generation sequencing technology, has comprehensively characterized the genome, transcriptome, and miRNome of 104 Taiwanese colorectal cancer tumor tissues, thereby establishing a robust multi-omics database specific to colorectal cancer. Beyond the creation of this cancer tissue database, we also employed next-generation sequencing to establish a liquid biopsy database. This includes the extracellular vesicle miRNome from 290 metastatic colorectal cancer samples, referred to as the blood EV-miRome. The team leveraged advanced bioinformatics tools, clinically relevant statistical analyses, and cross-referencing methodologies to pinpoint multifaceted biomarkers. These biomarkers not only offer an in-depth perspective on the progression dynamics of metastatic colorectal cancer but also serve as tools for longitudinal therapeutic response monitoring and outcome prediction. Integrating the extracellular vesicle miRNome with functional genomics data from cancer tissues, we proposed that the miRNAs contained within extracellular vesicles, secreted by patients with distant metastasis, contribute to cellular dormancy during metastatic bloodstream invasion and subsequently reactivate the mesenchymal-epithelial transition (MET) mechanism upon colonization of distal metastatic sites. Finally, to probe the dynamic molecular interplay within the tumor microenvironment, we are also utilizing the state-of-the-art spatial transcriptome platform to further characterize the constituents and cellular networks related to tumor progression. Students participating in the team's pivotal projects will acquire fundamental skills in bioinformatics analysis, clinical statistical methods, and foundational molecular/cell biology techniques. They will identify potential therapeutic targets through bioinformatics, logically construct biological regulatory networks, and design experiments for validation. Furthermore, students will enhance their collaborative proficiency, hone their communication skills, and gain familiarity with laboratory protocols and operations.
