Epidermal growth factor receptor (EGFR) tyrosine kinase inhibitor-targeted therapy has been established to treat patients with lung adenocarcinoma harboring EGFR-activating mutations. Platinum-based chemotherapy is often used to treat patients with EGFR wild-type lung ADC, but the overall outcome and life quality of the patients are unsatisfactory. We apply quantitative tissue proteomics to establish differential protein profiles associated with cancer stage and EGFR gene mutations. Bioinformatics analysis allows us to select potential biomarkers for further validation. The functional characterizations are performed by using cancer cell lines. The success of this project will significantly advance the development of biomarkers to benefit patients and provide new insights into lung adenocarcinoma tumorigenesis.
